Effect of Cheongpesagan-tang on the Changes of the Weight, Plasma and UCP mRNA Expression in Obese SD Rats Induced High Fat Feed

Abstract
Objective: This experimental study was designed to investigate the effect of Cheongpesagan-tang extract on the obese SD rats induced high fat feed.

Material and Methods: The weight of the whole body, liver and adipose tissues, food consumption, uncoupling protein(UCP) mRNA and NPY expression, and plasma levels of total-cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, leptin, insulin, glucose, SGOT and SGPT were measured in obese rats administered high fat feed and Cheongpesagan-tang extract(0.25mg/g, daily) for 5weeks. These were then compared with those of control groups administered physiological saline. Statistical comparisons among the normal, control and sample group were done by anova multiple test and pearson correlation test.

Results: Body weight gain, food consumption and the weight of liver and dpididymal white adipose tissue were significantly decreased in sample group compared with control groups(P<0.05). The levels of triglyceride, LDL-cholesterol, serum leptin, insulin and glucose were significantly decreased in sample group compared with control groups(P<0.05). The both expressions of UCP1 m-RNA in BAT and UCP2 m-RNA in EWAT were up-regulated in sample groups compared with control groups. The expressions of NPY in ARC, PVN and DMN area of hypothalamus was scale-down in sample groups compared with control groups.

Conslusions: These results shows that Cheongpesagan-tang has an effects on the treatment of obesity and type 2diabetes. Further study about the receptor and resistance that of leptin and insulin, is still required.
